It has been well known that Mg-ATP is fairly specific for inducing the clearing response of actomyosin at physiological ionic strengths. Although inorganic pyrophosphate is shown to cause dissociation of actomyosin at high ionic strengths, it does not act on actomyosin at all at low ionic strengths. However, it was found in the present study that pyrophosphate could induce the clearing response of actomyosin in the presence of 0.15M KCl, 1mM MgCl_2 and 0.01M Tris buffer, pH7.2, provided that 1M ures or 7% formamide was also present. The presence of Mg ions was required for the action of pyrophosphate. Urea, in concentrations less than 0.7M, was ineffective. ADP was effective to a similar degree as pyrophosphate, but AMP or inorganic orthophosphate did not work at all. Under the influence of 1M urea the myosinaggregate was somewhat disaggregated so that the actomyosin cleared in the presence of pyrophosphate showed a smaller birefringence and a larger extinction angle than in the case of the usual ATP-induced clearing. The present work suggests that pyrophosphate or ADP causes clearing or dissociation of actomyosin at a low ionic strength, if the actinmyosin binding is somehow weakened by the so-called muscle relaxing agents-urea or formamide.
