<p>Development of tetraploids is a useful for breeding Catharanthus roseus in terms of flower enlargement and, imparting disease resistance and sterility. Some screening processes are needed to obtain tetraploids following polyploidization using spindle toxins such as colchicine, because plants treated with a spindle toxin often show ploidy chimeras. First, we investigated changes in ploidy level with growth of colchicine-treated plants. Ploidy level varied among primary branches in colchicine-treated plants, and the main axis showed mericlinal chimeras. However, ploidy levels were stable for periclinal chimeras in each primary branch. Plants developed from cuttings obtained from the same primary branch showed the same ploidy, suggesting that tetraploids could be obtained from cuttings of primary branches that showed 4x. We also tested a screening process by shoot regeneration from leaf segment culture. Two periclinal chimeras were used for the study. Ploidy level of regenerated shoots was the same as that of the L1 layer of the leaf segment from which it emerged. This result suggested that tetraploids could be obtained by shoot regeneration from leaf explants with an L1 layer showing 4x.</p>
