Our previous studies suggested that electrical stimulation with random pulse train (RdPT) waveform have a potential to enhance osteogenesis and mineralization of osteoblasts in vitro. However, the effect of RdPT on collagen crosslink formation, which contributes improving the mechanical properties of bone matrix created by the cells, have not investigated yet. The objective of this research is to investigate the effect of RdPT on collagen crosslink formation by osteoblasts in vitro. To this end, lysyl oxidase (LOX) enzyme activity of osteoblasts was analyzed and insolubility of collagen was evaluated by measuring hydroxyproline (HyP) concentration. MC3T3-E1 osteoblastic cells were stimulated electrically by two types of electrical stimulation pattern, periodic pulse train (PrPT) and RdPT. RdPT increased the percentage of insoluble collagen over cultured period comparing to PrPT and control. Additionally, RdPT also increased in LOX activity on day 21 when compared with control. These findings suggest that the random electrical stimulation pattern can enhance the mechanical strength of bone matrix through promoting intermolecular collagen crosslinking formation.
