Therapeutic angiogenesis using bone marrow cell transplantation has been reported, and it was found that this therapy is useful for arterio sclerosis obliterans(ASO). We attempted the same procedure using peripheral blood stem cell collection(PBSCC).1. Clinical Study One-hundred-and-nine patients were included in this study,770f which were hemodialysis patients. They were all peripheral arterial disease(PAD)patients(101, lower limbs.8, upper limbs). The patients were given 5μg/kg/day G-CSF for four days subcutaneously. PBSCC was conducted on day 4. For each patient,8.3Lof peripheral blood was treated with an apheresis machine(CS-3000 0r Spectra)and an average of 3.6x10'CD 34 cells were harvested with mononuclear cells, red blood cells and platelets. We did not purify the CD 34 cells using the magnetic beads method, and the CD 34-enriched fluid(56.5ml)was injected at 113 points of the limb muscle on the same day. Thirty patients recovered. The disappearance of foot ulcers continued for four years. The symptoms of another 36 patients did not change but their limbs were saved. The other 43 patients had amputations because the ulcers were necrotic and infected. II. Experimental Study:We made an ischemic model using beagles(8-10 kg)and conducted a case-control study.
