Loss of ε-Aminocaproate Metabolism in Corynebacterium aurantiacum

Corynebacterium aurantiacum B-2 is characterized by its ability to grow on ε-caprolactam and cyclic oligomers of ε-aminocaproic acid (ACA) larger than the trimer as the sole source of carbon and nitrogen. Slow growth of B-2 colonies on an ACA cyclic tetramer agar medium kept in a cold room (12±3℃) for 5 weeks resulted in production, with a 32% frequency, of cells of a metabolic variant which could not grow on the cyclic tetramer, ε-caprolactam or ACA. This variant strain retained hydrolysis activities toward the cyclic tetramer, the linear dimer and ε-caprolactam, but lost the ACA transaminase activity. This high rate of curing of the ACA metabolism and the lack of reversion to the original characteristics suggest that the gene for ACA transaminase may be carried by a plasmid.
