Description
Fluorescent dyes were examined for staining nucleus in protoplasts from soybean suspension cultures and lettuce cotyledons in vital state. For this purpose, protoplasts were stained with rhodamine 6G, Hoechst 33342 and acridine orange. For comparison, DAPI was used for fixed protoplasts. Hoechst 33342 could be effectively used at very low concentrations for both protoplasts, so very useful for studying fusion process of plant protoplasts. Other dyes could be used for staining nucleus of soybean protoplasts but not good for lettuce protoplasts.
