Inhibitory effects of baicalein and wogonin on the growth of B16 melanoma cells

We examined the effects of two flavones, baicalein and wogonin, on the growth and differentiation of mouse B16 melanoma cells in culture. Baicalein and wogonin are plant flavones isolated from the processed root of scutellaria baicalensis Georgi, and have similar chemical structures. Although, these two flavones inhibited the growth of melanoma cells in a dose-dependent manner, the growth inhibitory effect of baicalein was greater than that of wogonin. Both baicalein and wogonin accumulated in the cells in the G0/G1 phase. However, treatment with wogonin led to drastic changes in cell shape, cytoskeleton, and subcellular organelles, whereas no significant morphological changes were observed in cells treated with baicalein. Treatment with wogonin also induced changes in cell function such as melanogenesis, cell detachment, and enzyme activity. These findings indicate that the chemical structure and the antitumor activity of baicalein and wogonin are similar, but their action mechanisms differ.
