Description

We developed a highly sensitive technique using high-performance liquid chromatography (HPLC) to determine the amount of hexosamine. This method is dozens of times more sensitive than conventional colorimetric determination methods. The amount of hexosamine in the acid hydrolyzate of an extract of stomach specimen showed a close correlation with the amount of gastric mucin. Combining these two methods, we determined the hexosamine content in each layer of rat gastric corpus and antral mucosa previously treated with aspirin or a synthetic prostaglandin E_2 derivative, Arbaprostil, and obtained the following results. i) The amount of hexosamine in both the mucus gel layer and the mucosal layer was reduced by the administration of aspirin. ii) The administration of Arbaprostil did not elicit a change in the amount of mucin in the mucus gel layer. Furthermore, we used this HPLC method to determine the mucin content in human gastric specimen obtained by biopsy. A considerably greater amount of mucin was observed in the region of the lesser curvature of the angle than in other regions.
