cDNA Cloning and Mutagenesis Study of Liver-Type Prostaglandin F Synthase, and Identification of the Prostaglandin F Producing Cells in the Liver

Prostaglandin F synthase (PGFS) is a multi-functional enzyme, and catalyzes the reduction of prostaglandin (PG)D2to 9a, 11f -PGF2and that of PGH2to PGF2aon the same molecule. PGFS has at least two isoformes, the lung-type enzyme’ (Km value of 120 µM for PGD2) and the liver-type one2(Km value of 10 µM for PGD2). The bovine lung-type PGFS is a:monomeric protein with a Mr of 36,666 consisting of 323 amino acids, and its amino acid sequence shows high homology compared with those of other aldo-keto reductase family (AKR family) members. However, the liver-type PGFS cDNA has not been isolated, and the primary structure has not been known. In the present study, we decided the primary structure of bovine liver-type PGFS and deduced the amino acids related the affinity for PGD2by the mutagenesis study.3Furthermore, our immunohistochemical study revealed PGF-producing cells in the liver.4
