Abstract 1. 1. Action potentials of smooth muscle cells of the guinea pig ureter were recorded by sucrose gap method, and the membrane potential was changed by applying polarizing current through a suction electrode. 2. 2. Depolarization reduced the amplitude and the maximum rate of rise of spikes, whereas hyperpolarization increased them. 3. 3. The initial spike in a train of discharges was influenced most remarkably by change in membrane potential. 4. 4. The relationship between membrane potential and maximum rate of rise of spike could be expressed by a sigmoid curve. Increase of Ca concentration shifted the curve upwards in the hyperpolarized range. 5. 5. The role of Ca in the transfer of charge responsible for the spike potential is discussed.
