A Simple Technic for Light and Electron Microscopic Observation of the Same Sections: Study of Human Hyperplastic Lymph Nodes

Semithin sections (1,500 to 1,800A thick) were mounted on copper grids and stained with a solution of basic fuchsin and methylene blue. After being examined with an optical microscope and photographed, sections were further stained with uranyl acetate and lead for electron microscopic observation. This simple technic provided precise orientation and ultrastructural details of particular cells previously spotted by light microscopy. Sections from two cases of human hyperplastic lymph nodes were examined by this method. Rare atypical histiocytes containing Langerhans cell granules in one case, and foreign-body giant cells containing a filamentous layer at the periphery in the other case, were fully examined by both light and electron microscopy by using the same grids.
