The fine glass capillary electrode technique has been developed to measure the corneal potential of the enucleated whole eye. The trans-epithelial and trans-endothelial potentials were a few mV, and their magnitude and polarity were dependent on the composition of bathing solutions and temperature. The trans-endothelial potential was 1 mV or less, and the stromal side was always negative with reference to the aqueous humour. The corneal potentials measured in different parts of the cornea were not identical, showing that the stroma was not in equi-potential. The potential difference of concentration gradient established across the epithelium was in agreement with the Nernst equation of diffusion potential in some range of concentration gradient. At the interface of the denuded stroma, a boundary potential developed that was always negative in the stroma side with reference to the bathing solution.
