Microfabricated directional coupler-based biosensor for afinity sensing

A directional coupler (DC) — based biochemical sensor was developed for affinity sensing. The DC sensor consisted of two parallel waveguides with a sensing area that can be miniaturized to several tens of μm. Values experimentally obtained with water and ethanol agreed well with those obtained by simulation. Reproducible changes were obtained by repetitive refractive index changes with air, water, and ethanol. The DC sensor showed reproducible responses, and the distribution of the response was within 5 % with respect to the maximum signal difference. Real time detection of streptavidin (SA) was conducted with biotin immobilized on the DC surface. With a 616-μm long DC, achieved detection limit was 11.3 μg/ml.
