Abstract 1. 1.|Using purified samples of laccase from latex, it was demonstrated that their copper content was quantitatively proportional to the absorption band at 615 mμ. The extinction coefficient at 615 mμ, which is attributable to the cupric atoms contained in the enzyme, was estimated to be 1.1 · 103/cm/gram atom of copper/liter. 2. 2.|It was shown that the oxidized (cupric) enzyme is reduced by the substrate (hydroquinone or ascorbic acid) under anaerobic conditions, and the reduced (cuprous) enzyme is oxidized by molecular oxygen. By investigating these processes stoichiometrically, it was concluded that the reaction catalyzed by laccase involves the following two steps: 2Cu++ + hydroquinone → 2Cu+ + p-quinone + 2H+ 2 Cu + + 1 2 O 2 + 2 H + → 2 Cu ++ + H 2 O .
