抄録

We investigated the availability of a recentdeveloped nucleic acid stain, SYBR Green I, bacterial counting method. Comparison of bacterial counts made with SYBR Green I and DAPI method was conducted using coastal seawater samples. Bacterial counts with both methods were significantly correlated (r=0.60, n=23). However, the ratio of bacterial counts with SYBR Green I and DAPI method was 2.0±0.5, indicating that the count with SYBR green I method was about twice higher than with DAPI method. Bacteria were intensively stained with SYBR Green I and easier to observe under epifluorescence microscopy than with DAPI method. Although it is difficult to identify the reason for difference of the bacterial counts, our results imply a possible existence of bacteria which cannot be detected with the DAPI method used previously.
