To recover functional genes from specimens of extinct or endangered species is of prime importance from the view point of conservation of genetic resources as well as that of evolutional studies of genes. Hides from stuffed specimens or pelts are expected to serve as suitable materials for the purpose. However, methods have to be developed so that the damages to the original precious specimens will be minimal. Methods so far described in the literature requires relatively large amounts of the specimens and not suitable for the analysis of highly valuable specimens. In the present paper, an improved method is described to extract genomic DNA from the hide or pelt specimens of approximately 1mm2. Parameters and conditions optimal for PCR analysis of the extracted DNA are also investigated. Employing the procedures we established, genomic DNA was extracted from various carnivoran hide specimens with different passage of time after their preparation, and the partial DNA sequences of the genes coding for amelogenin, a major protein component of enamel of the teeth, were determined.
