Comparative in vitro evaluation was made between rifampicin and rifamycin SV. Minimum inhibitory concentrations were estimated by two-fold agar-plate dilution method. Each test strain was cultured overnight in Trypto-soy broth, one loopful of which was streaked on test agar-plate, and MICs were determined after 24 hours incubation at 37°C.<BR>Rifampicin showed very high activity against GRAM-positive bacteria, and was also moderately effective against many species of GRAM-negative rods, whereas rifamycin SV was confirmed to be less active against GRAM-positive bacteria and quite inactive against GRAM-negative species.<BR>One hundred clinical isolates of Staph. aureus, including multiple-resistant strains, were completely suppressed below 0.006mcg/ml of rifampicin, the activity being higher by far than other antibiotics. However, against strains of Shigella and E. coli, MICs of rifampicin ranged 6.25-42.5 mcg/ml, less active as compared with those of chloramphenicol or tetracycline. Among Proteus, Klebsiella and Pseudomonas, strains of Proteae were most susceptible to rifampicin, the MICs being ranged below 25 mcg/ ml.<BR>Changes in various experimental conditions of in vitro testing such as kinds and reactions of the test media, and the sizes of inocula did not cause any noticeable fluctuation of MIC.<BR>These observations as well as the experimental data on the absorption and tissue distributions of rifampicin strongly suggest that this substance may be highly effective against infections due to GRAM-positive bacteria, and also useful for the infections due to GRAM-negative rods.
