Description
Detection of Legionella species using the loop-mediated isothermal amplification (LAMP) method was investigated with the “Loopamp Legionella detection reagents kit E” (Eiken Chemical Co., Ltd.). The detection limit of the LAMP method was 3.7×101 CFU/test, approximately equivalent to the limit claimed by the instructions (60 CFU/test). The LAMP method detected positive reactions for 17 of 100 Legionella samples which gave negative reactions by the culture method, indicating that the sensitivity of the LAMP method was higher than the culture method. No false positive results were obtained in the screening of 5 organisms other than Legionella species. All visual results were equal to those obtained using a turbidity meter. The LAMP method can specifically detect Legionella species at levels lower than the detection limit of the conventional culture method in a short time, so is useful for the prevention of nosocomial Legionella infection.
