Abstract

We discovered that a mutant defective in vacuolar functions (slp 1-1 mutant) leaked glutathione, but strain KL-197-12 B (slp 1-1 mutant) had weak fermentation. Therefore, hybrids were bred by repeated crossings between a temporal parent KL-197-12 B and a repetitive parent Kyou-kai No. 7 (K-7). Strain KL-197-12 B (α) was crossed with haploids of strain K-7 to yield first generation-haploids (1 HY). The haploids selected from 1 HY were back-crossed with haploids of K-7 to yield second generation-hybrids (2 HY), and the same process was continued until the strains had restored fermentation. Several strains were comparable to K-7 on fermentation of sake mash at a laboratory scale. Strain (No. 133) was most suitable in glutathione-leakage and fermentation.
