This study was undertaken to examine effects of the time and temperature of exposure to dimethyl sulphoxide (DMSO) prior to freezing on survival of 8-cell rat embryos after freezing and thawing. Embryos were cooled to-79°C at 0.5°C/min and thawed in air at room temperature. Survival of frozen-thawed 8-cell rat embryos was assessed by their ability to develop into blastocysts during culture. Results are as follows. 1. An exposure to 1.5M DMSO at 30°C for 0.5min prior to freezing conferred higher (P<0.05) survival of frozen-thawed embryos than an exposure at 0°C for the same time. 2. About 42% of embryos survived freezing after only a 0.1min exposure to 1.5M DMSO at 0°C prior to freezing, almost the similar percentage that survived after 5-90 min exposures. To the contrary, a 120min exposure to 1.5M DMSO at 0°C prior to freezing decreased the survival rate of frozen-thawed embryos in comparison with those of embryos frozen after 0.1-90min exposures, although differences were not significant. 3. Regardless of the DMSO concentrations(0.8-2.5M), an exposure of 0.1 and 15min to DMSO at 0°C prior to freezing conferred almost the same protection against freezing damage. In conclusion., DMSO permeation enhances survival of frozen-thawed 8-cell rat embryos, but it is not the sole factor that determines their survival.
