N-Butyl-N',N'-dimethyl-N-nitrosourea (DM-BNU), the spontaneous (hydrolytic) activation rate of which is 4×10^<-4> times that of N-butyl-N-nitrosourea (BNU), is metabolized to N-butyl-N'-methyl-N-nitrosourea (MM-BNU) in the presence of S9 mix. It is, therefore, suggested that the carcinogenicity of DM-BNU, which is as potent as that of BNU, may be attributed partly to the demethylated MM-BNU, which is more readily activated to a carcinogenic reactant. This hypothesis is supported by the mutagenicilies of these carcinogens in E. coli in the presence and absence of S9 mix.
